چکیده

Fifty-five-day-old Sprague-Dawley female rats were given an i.v. injection of 5 mg 7,12-dimethylbenz(a)anthracene, and 15 to 30 days after the appearance of pal pable mammary tumors the rats were given (a) an estradici benzoate (EB) implant in the median eminence (ME), (b) an EB implant outside the ME, (c) an EB implant in the cerebral cortex, or (d) a cholesterol implant in the ME. At 25 days after implantation, the average number of tumors per rat and percentage of increase in size and total weight of tumors in rats with an EB implant in the ME were significantly greater than in all other groups. This is attributed to the observed increase in serum prolactin levels as a result of the direct action of EB on the hypothalamus, pituitary, or both.

Effects of Alterations in Early Hormonal Environment on Development and Hormone Dependency of Carcinogen-induced Mammary Tumors in Rats1

The purpose of this study was to determine the early role of estrogen and prolactin on subsequent hormone dependency of carcinogen-induced mammary tumors. Virgin 57-day-old Sprague-Dawley rats were given i.v. injections of 7,12-dimethylbenz(a)anthracene(DMBA). One day prior to and 7 days after DMBA administration, the rats were divided into separate groups and given: a daily 0.1-ml s.c. injecti...

The nature of estrogen and prolactin effect on mammary tumorigenesis.

Increased release of pituitary prolactin as a result of placement of electrolytic lesions in the median eminence of the hypothalamus greatly accelerated the growth rate of carcinogen-induced mammary tumors in rats. Ovariectomy in these rats induced a rapid regression of tumors in spite of high plasma levels of prolactin. Grafting a pair of ovaries into ovariectomized rats with lesions of the me...

Effects of High Dietary Fat on the Growth and Development of Ovarian-independent Carcinogen-induced Mammary Tumors in Rats1

This study examined the influence of high dietary fat intake on the development of ovarian-independent mammary tumors in both vehicle-treated controls and rats made deficient in estrogen and prolactin during tumor induction. The majority of 7,12dimethylbenz(a)anthracene(DMBA)-induced mammary tumors in rats are dependent on estrogen and prolactin for growth, and suppression of prolactin and estr...

Investigation of p53 and p27 expressions in the N-nitroso-N-methylureainduced breast cancer in female Wistar Albino rats

Introduction: N-nitroso-N-methylurea (NMU) is a carcinogen from nitrosamines family, which has been used to induce breast cancer in rodents. This model of breast cancer is very similar to the estrogen dependent breast cancer in human. As a continuation of our recent works, in the present study, the expressions of both p53 and p27 were investigated in NMU-induced breast cancer in Wistar Albin...
